Help & Instructions
1. Data Format
Your Excel/CSV file should have:
- First column: Cycle numbers (1, 2, 3, ...)
- Subsequent columns: Fluorescence values for each well (A01, A02, B01, etc.)
2. How to Use
- Load your data or use the built-in demo
- Click wells in the 96-well plate grid to select/deselect
- 'Single/Multi-well Analysis' shows Left=Processed (filter+smooth+normalize+threshold+Ct) and Right=Raw
- 'Batch Run' analyzes all wells at once
3. Processing Pipeline
- Filter: Outliers replaced by local median (3-sigma rule)
- Smooth: Savitzky-Golay filter (poly=3, window=5) — preserves peak shape
- Normalize: Baseline mean subtraction
- Threshold: Manual OR baseline_mean + n * baseline_sd
- Ct: Linear interpolation at threshold crossing
