Help & Instructions

1. Data Format

Your Excel/CSV file should have:

  • First column: Cycle numbers (1, 2, 3, ...)
  • Subsequent columns: Fluorescence values for each well (A01, A02, B01, etc.)

2. How to Use
  • Load your data or use the built-in demo
  • Click wells in the 96-well plate grid to select/deselect
  • 'Single/Multi-well Analysis' shows Left=Processed (filter+smooth+normalize+threshold+Ct) and Right=Raw
  • 'Batch Run' analyzes all wells at once

3. Processing Pipeline
  • Filter: Outliers replaced by local median (3-sigma rule)
  • Smooth: Savitzky-Golay filter (poly=3, window=5) — preserves peak shape
  • Normalize: Baseline mean subtraction
  • Threshold: Manual OR baseline_mean + n * baseline_sd
  • Ct: Linear interpolation at threshold crossing
